Isolation and screeningfor thermophilic lignin -degrading actinomycetesfrom humus samples collected at the Bai Bang Paper Company

ABSTRACT
Lignin is a natural and complex compound that is recalcitrant to degradation and must be removed during pulp and paper production. In Vietnam, the materials for pulp and paper production are Acacia and Eucalyptus wood, with the lignin content of about 27.6%. At present, environment-friendly biopulping technology exploits lignin-degrading enzymes of microbial origin to improve product quality and minimize water pollution. In this paper, from 20 samples of humus samples collected at the Bai Bang Paper company, 47 lignin degrading thermophilic actinomyceteswere isolated. Among the isolates, the isolated CX9strainwas one of the species which showed remarkably high activity of Lignin peroxidase (7345 U/L). The optimal temperature for the growth of CX9 was at 37-45oC. On the ISP2 medium, the aerial mycelia were grey and branched, and purple pigment was secreted. The results of morphological study and phylogenetic analysis of 16S rDNA gene suggested that the isolated CX9 was close totheStreptomycesthermoviolaceus, hence named as StreptomycesthermoviolaceusCX9.
